FAQs
We recommend that you use the liquid culture to inoculate grains and use the grain spawn to inoculate your growth substrate. Whether it is sawdust, hay or cardboard. Just break up the grain spawn once the mycelium have taken over it. Then inoculate the growth substrate you want to use.
How to inoculate an agar plate with liquid culture? ›
Using a sterile pipette tip or toothpick, select a single colony from your LB agar plate. Drop the tip or toothpick into the liquid LB + antibiotic and swirl. Loosely cover the culture with sterile aluminum foil or a cap that is not air tight. Incubate bacterial culture at 37°C for 12-18 hr in a shaking incubator.
How do you inoculate a liquid bacterial culture protocol? ›
Procedure
- Add 1000X kanamycin to the LB so that the final concentration is 1X. Swirl to mix.
- Using a pipette tip, touch a single colony on the plate.
- Touch the pipette tip to the LB in the bacterial culture tube and lightly swirl. At least one cell must remain in the media.
- Grow at 37°C overnight while shaking.
Can you inoculate logs with liquid culture? ›
Once sterilized and cooled, the plugs can be inoculated with mushroom culture using either grain spawn, agar wedges or liquid culture.
Can you inoculate liquid culture from glycerol stock? ›
A small amount of sample from the glycerol stock is streaked onto a freshly prepared agar plate to be cultivated for further analysis. Inoculate an overnight liquid culture. Create Bacterial Glycerol Stock (BGS). Transfer BGS on a screwcap cryogenic tube and store in a -80°C freezer.
Can you put agar into liquid culture? ›
Culture media are usually made in solid form (agar) or liquid form (liquid culture or broth). Agar nutrient mixtures are typically the same as broth with agar added to solidify the medium.
Is LC faster than agar? ›
If you're a mushroom cultivator ready to level up your growing game, then it's time to consider liquid culture. Liquid culture offers faster colonization and more predictable outcomes than spore inoculation, and an easier process than agar work.
How long does liquid culture inoculation take? ›
This should take 10-14 days at warmer temperatures, or 16-21 days at temperatures around 70 degrees. Please research the specific species of mushroom you are cultivating in order to incubate at correct temperature.
Can you inject spores directly into liquid culture? ›
To inoculate a liquid medium for mushroom liquid culture, you can use various sources of mushroom mycelium: Spores: Spores are a common choice for inoculation. They can be obtained from a mature mushroom's spore print and introduced directly into the liquid medium using a sterile scalpel.
How do you store liquid culture after inoculation? ›
Store in a cool, dark place: The liquid culture should be stored in a cool, dark place such as a refrigerator or a cool basem*nt. This will help to slow down the growth of the mycelium and extend the shelf life of the culture. Preventing UV light will also keep the mycelium viable for longer.
Using a sterile inoculation loop, select a single colony from your bacteria streaked LB agar plate. Dip the inoculation loop into the liquid LB and swirl. Discard the inoculation loop. Loosely cover the culture with sterile aluminium foil or a cap that is not air tight as bacteria needs air.
How to inoculate agar plates? ›
Inoculating a Plate from a Broth Culture
- Sterilize the inoculating loop.
- Remove the cap from tube. ...
- Flame the lip of the tube.
- Place sterile portion of inoculating loop into broth, then remove.
- Flame the lip of the tube.
- Replace the cap.
- Gently streak the surface of an agar plate with the inoculating loop.
What is the difference between agar plates and liquid culture? ›
Agar dishes are believed to be more trustworthy and accurate, but they can be more difficult to handle than liquid cultures. To develop your mushroom cultivation skills, it is advisable, to begin with liquid cultures and then on to agar dishes.
Does mycelium grow in liquid culture? ›
Liquid culture is a nutrient-rich solution used to grow mushroom mycelium. It's a mix of mushroom mycelium and a sterilized liquid, often used to inoculate grain to create grain spawn, which is essential for mushroom growing.
Can you go from liquid culture to agar? ›
To transfer liquid culture to agar, you'll be using a syringe. You'll liquid culture will either already be in a syringe, or it will be in a jar that you'll first need to pull into a syringe. Either way, you'll need to agitate your liquid culture to break it up, so that each drop of liquid contains a little mycelium.
Is liquid culture the same as spore syringe? ›
Unlike a spore syringe that contains dormant spores, a liquid culture contains living, active mycelium. The liquid culture solution is a nutrient-rich liquid that provides the mycelium with the sustenance it needs for faster colonization.
